猪APOA5和APOC3基因mRNA的表达差异及与肌内脂肪沉积的相关性分析

摘    要:为了探究载脂蛋白A5(apolipoprotein A5,APOA5)基因和载脂蛋白C3(apolipoprotein C3,APOC3)基因组织表达差异及其与肌内脂肪(intramuscular fat,IMF)沉积的相关性,本研究利用荧光定量PCR方法分析APOA5和APOC3基因mRNA在可乐猪和江口萝卜猪7个组织中的相对表达量及其与IMF沉积的相关性。研究结果表明:APOA5和APOC3基因在可乐猪和江口萝卜猪的7个组织中均有不同程度表达,在肝脏中表达量最高,在皮下脂肪和背最长肌中表达量次之;APOA5基因在可乐猪多数组织中的表达量显著低于江口萝卜猪(p〈0.05),而APOC3基因未呈现类似规律性;可乐猪中,除心脏外,其它组织均表现为APOC3基因mRNA表达量高于APOA5,江口萝卜猪心脏、肝脏、脾脏、小肠和背最长肌中同样呈现上述表达差异。相关及回归分析表明,IMF沉积量与可乐猪APOC3 mRNA表达丰度呈线性正相关(p〈0.05)。根据研究结果,可以推测APOA5和APOC3基因与IMF沉积有一定关联性。

Abstract:To explore the expression level difference and relationship with intramuscular fat (IMF) deposition, relative expression of apolipoprotein A5 (A POA 5) and apolipoprotein C3 (A POC3) genes in seven tissues of Kele pig and Jangkou Luobo pig were analyzed by real-time fluorescence quantitative PCR. The resuks showed that A POA 5 and A POC3 genes expressed in the whole seven tissues with distinguishing expression levels. Expression quantity of the two genes were the highest in liver, followed by the adipose and longissimus muscle. For A POA 5 gene, its expressions in Kele pig were significantly lower than Jiangkou Luobo pig in the most tissues (p〈0.05), while the A POC3 gene did not show the similar regularity. The expression quantity of A POC3 gone was higher than A POA 5 in almost tissues expect for heart in Kele pig, and in Jiangkou Luobo pig the tissues of heart, liver, spleen, intestine and longissimus muscle showed the same results. Analysis the correlation of gene expression quantity of APOA5 and A POC3 genes and content of IMF, the results indicated that the level of IMF were linear dependence with the gene expression quantity of APOC3 mRNA in Kele pigs significantly (p〈0.05). According to the report and presumably A POA 5 and A POC3 genes may be related to IMF deposition.
