蝴蝶兰叶片诱导类原球茎初探

摘    要:以蝴蝶兰杂交品种H807无菌苗的叶片为材料,研究了防褐化物质(柠檬酸、抗坏血酸及活性炭)和细胞分裂素(6-BA与TDZ)对类原球茎诱导的影响。结果表明:在添加70mg/L抗坏血酸的培养基中,叶片褐化级别仅为1级,防褐化效果最佳;3mg/L的TDZ对蝴蝶兰叶片诱导类原球茎的诱导率最高,为75.0%。在以1/2MS附加70 mg/L抗坏血酸的基础上,激素组合TDZ3mg/L+NAA 1mg/L对蝴蝶兰H807类原球茎的诱导效果最佳。

Abstract:There are serious browning and low induction rate when the protocorn-like body(PLB) was induced from leaves of Phalaenopsis amabilis.Using the grem free leaf of Phalaenopsis amabilis H807 as materials,the effects of anti-browning materials(citric acid,ascorbic acid and activated charcoal)and cytokinins(6-BA and TDZ) on the protocorn-like body induction was investigated.The results indicated that the rank of anti-browning only reached the level 1 with 70 mg/L ascorbic acid,which was best to prevent in vitro leaves from browning.The protocorn-like body induction rate reached 75.0% with 3 mg/L TDZ.On the medium with 1/2MS+70 mg/L ascorbic acid,hormone combination of 3 mg/L TDZ+1 mg/L NAA was the best for PLB induction.
