摘    要:旨在研究甘肃省天水马铃薯-小麦轮作模式下马铃薯根内微生物多样性的变化规律。以马铃薯单一连续种植模式为对照,利用Illumina测序[细菌16s rRNA(V3-V4)和真菌ITS1]分析马铃薯根内微生物的多样性及组成。结果显示,与连作对照相比,轮作模式下马铃薯根内微生物的多样性、群落组成及物种丰度均有明显差异,且在马铃薯块茎膨大期,根内益生菌Pseudomonadaceae、MortierellaFlavobacterium等的丰度水平明显提高。由此得出,耕作模式影响马铃薯根内微生物多样性的特征,并且推断轮作模式下形成的根内微生物群落更有利于马铃薯的生长和抗逆性的提高。此外,对环境因子与马铃薯根内微生物进行关联性分析,结果显示,马铃薯根内微生物与环境因子有显著的相关性(尤其土壤pH、含水量、铵态氮等因素)。上述研究结果揭示西北旱区耕作模式和环境因子对马铃薯根内微生物多样性及群落组成具有显著影响,该结论对西北旱区马铃薯的绿色高效生产可资借鉴。

Abstract:This experiment was conducted to study the microbial diversity in potato root under the tillage pattern of potato-wheat rotation in Tianshui of Gansu province. As a control,the microbial diversity in potato root under the tillage pattern of single continuous potato planting was studied. Illumina sequencing (bacterial 16s rRNA (V3-V4) and fungal ITS1) was used to analyze the microbial diversity and composition in potato root. The results showed that the diversity,community composition and species abundance of the microorganisms in potato roots grown in potato-wheat rotating field were significantly different from those in single continuous potato planting field. Notably,the abundance levels of the root probiotics Pseudomonadaceae,Mortierella and Flavobacterium were significantly increased during the stage of potato tuber expansion stage in potato-wheat rotating field.In conclusion tillage patterns affect the potato root microbial diversity,and we can infer that the root microbial community formed under rotation pattern is more conducive to potato growth and stress resistance. In addition,we conducted a correlation analysis between environmental factors and potato root microorganisms,and the results showed that potato root microorganisms were significantly correlated with environmental factors (especially soil pH,water content,ammonium nitrogen and other factors). This study revealed that the typical potato cultivation mode in northwestern China and environmental factors affect the potato root microbial diversity and community composition,which provides important fundamental theory which can facilitate the development of potato sustainable and green production in the semi-arid area of Northwestern China.
