摘    要:采用SDS法、热CTAB法、高盐低pH法和改进CTAB法分别提取高羊茅(Festuca arundinoceaSchreb.)、草地早熟禾(Poa pratensis L.)和多年生黑麦草(Lolium perenne L.)3种冷季型草坪草基因组DNA.结果表明,改进CTAB法为最佳的提取方法,分离的DNA产率高、纯度高、质量高,经纯化后电泳检测带型清晰.RAPD扩增显示,用该快速、简便、有效的方法提取的基因组DNA能扩增出清晰易辨的带型.

Abstract:The genomic DNA of three kinds of cool-season turfgrass (tall fescue,kentucky bluegrass and perennial ryegrass) was isolated by SDS method,hot CTAB method,high-salt-low-pH method and modified CTAB method. The results indicated that the modified CTAB method was the most effective. High yield ratio,high purity and good quality of the genomic DNA were acquired by the modified CTAB method.Clear DNA electrophoresis bands of purified DNA were displayed. RAPD amplification showed that clear and distinct DNA bands were amplified from the genomic DNA isolated by the modified CTAB method,which is a rapid,simple and effective method.
