哺乳期灌服亚精胺或精胺对早期断奶大鼠生长及肠道发育的影响

摘    要:为考察断奶前多胺诱导的肠道成熟是否具有缓解早期断奶应激的作用,本研究选取同窝14d清洁级SD哺乳大鼠分别经胃管连续3d灌服5μmol亚精胺,5μmol精胺或生理盐水(对照组)。17d实施早期断奶,一半幼鼠断奶后立即致死,剩余一半独立饲养至20d后致死,测定幼鼠体重及肠道发育指标。结果表明:亚精胺和精胺促进了17d哺乳大鼠肠道成熟,表现为肠道重量、肠长、小肠蛋白质、蔗糖酶活性的升高。亚精胺和精胺可有效阻止早期断奶失重,并降低或消除了早期断奶肠道的应激反应,表现为肠道重量、肠长、肠道蛋白质、肠道DNA和蔗糖酶、乳糖酶活性在断奶前后不受影响。亚精胺和精胺对肠道杯状细胞类型有显著影响,断奶后酸性杯状细胞分别增加20.1%(P>0.05)和89.5%(P<0.05)。综上可得出,断奶前灌服亚精胺或精胺具有促进哺乳期肠道成熟,降低早期断奶应激和增强黏液屏障的作用。

Abstract:To explore whether polyamine induced gut maturation could modify early-weaning tress, SD sucking rats of 14d old from same litter were infused 5μmol spermidine(SPD), 5μmol spermine (SPM) or saline (control) separately for 3 days. Pups were early-weaned on 17 d old, half were decapitated after weaning immediately, the residue were fed alone and decapitated on 20 d old. Body weight and gut development parameters were examined. SPD and SPM promoted 17d pup's gut maturation with higher gut weight, gut length, gut protein content, DNA content and sucrase specific activity. Meanwhile, SPD and SPM decreased body loss of early-weaning, and diminished early-weaning stress in pup's gut with no change for gut weight, gut length, gut protein content, DNA content and sucrase specific activity after weaning. Subtypes of goblet cell were significant influenced, SPD and SPM promoted acidic goblet cell by 20.1%(P > 0.05) and 89.5%(P < 0.05),separately. It suggested that infusion spermidine or spermine in suckling rats would promote gut maturation and then diminished early-weaning stress, enhance gut mucus barrier.
