茶树愈伤组织培养及其儿茶素累积

摘    要:对不同培养条件下茶愈伤组织生长及其儿茶素累积情况进行了研究,结果表明,激素 IBA 或KT 在单独使用时,效果各异,但两者间存在明显的互相作用效应,其中以 KT 10mg/L 和 IBA0.1mg/L 组合,对儿茶素累积最为有利,MS 培养基适合愈伤组织生长,而 White 培养基上的细胞儿茶素累积最高;提高蔗糖浓度有利于次生代谢产物的形成,但浓度过高不利于愈伤组织生长;在一个生长周期中,儿茶素累积的增加与愈伤组织鲜重增长密切相关,儿茶素含量在直线期末、静止期初最高。通过对培养材料来源的选择和培养基组成的调节,使茶愈伤组织儿茶素含量从不足1mg/gFW 提高至16—20mg/gFw。

Abstract:The growth and catechin accumulation of tea callus were investigated underdifferent culturing conditions.Results showed that the effects of IBA and KTwere different when they were used individuallg,but an apparent cooperationexisted between them when they were used together.The combination ofIBA 0.1mg/L and KT 10mg/L was the best for the catechin accumulation.MS basal medium was the best for callus growth and White basal mediumwas the best for the catechin accumulation in the callus.Increasing of thesucrose concentration was beneficial to the secondary metabolism,but thefresh weight of the callus would decrease when the sucrose concentrationwas too high.During a growth cycle,the accumulation of catechin wasclosely related to the increasing of the callus fresh weight.The catechincontent reached the highest between the late linear stage and the earlystationary stage.The catechin content of the callus was increased from below1 mg/g FW to 16-20 mg/g FW by adjusting the medium compositions and bychoosing the right materials for the cultures.
