苹果多聚半乳糖醛酸酶抑制蛋白的分离纯化

摘    要:以膨大期的富士苹果果皮为材料,通过匀浆、硫酸铵沉淀、透析、聚乙二醇6000浓缩、CM-SephadexC-50离子交换柱和Sephadex G-100分子筛凝胶过滤层析分离纯化出苹果多聚半乳糖醛酸酶抑制蛋白。用还原糖法测定其具有抑制多聚半乳糖醛酸酶活性,bradford法进行定量,经非连续SDS-PAGE分析为电泳纯,根据标准蛋白鉴定其亚基的分子量为41 kD。为苹果PGIP的纯化建立了一种有效的方法。

Abstract:Polygalacturonase-inhibiting protein(PGIP) was purified from extracts of apple cortical tissue by ammonium sulfate precipetation,ion-exchange chrom atography and gel filtration.PGIP activity was messured by determing reducing groups using the dinitrosalicylic acid procedure and PGIP weight was determined by bradford.The purified PGIP showed a single protein band during SDS-PAGE.The apple PGIP subnit has a molecular weight(as deternined by SDSPAGE) about 41kD.This study presented an effective approach for PGIP purification in Malus demestica and provided an experimental basis for the comparative study.
