摘    要:[目的]建立反相高效液相色谱法(RP-HPLC)测定芦丁中槲皮素含量的方法。[方法]用C18(250 mm×4.6 mm)色谱柱,流动相为水-乙腈-冰乙酸(1 060∶350∶45,V∶V∶V),流速为1.0 ml/min,检测波长为254 nm。[结果]采用水-乙腈-冰乙酸系统为流动相,分离效果好,峰型好,分离时间短,计算结果准确;以甲醇为溶媒提取时间短,提取杂质少。槲皮素含量在4~12 mg/L范围内呈良好的线性关系,回归方程Y=1 285X+3 645,平均回收率为99%。所测样品芦丁中槲皮素的含量平均为0.813 g/ml。[结论]采用RP-HPLC测定芦丁中槲皮素含量简便、灵敏、准确、重复性好,可作为质量和生产控制的有效方法。

Abstract:[Objective] The the content of quercetin in rutin,tested by means of the method of high performance liquid chromatography(RP-HPLC),was established.[Method] The testing condition was the size of the column C18 was 250 mm × 4.6 mm,the mobile phase was water-acetonitrile-acetic acid of 1 060∶350∶45(V∶V∶V),the flow rate was 1.0 ml/min and the detection wavelength was 254 nm.[Results] Under the condition of the water-acetonitrile-acetic acid system being taken as the mobile phase,the separation of peak was good and with good type;the separation time was short and the result was accurate.Under the condition of the methanol as the solvent,the extraction time was short and the content of impurity in extract was less.There was a good linear relationship while the range of the content of quercetin was 4-12 mg/L,and the regression equation was Y=1 285X+3 645,the average recovery was 99%.The average content of quercetin in tested sample was 0.813 g/ml.[Conclusion] The method was simple,sensitive,accurate and reproducible,which could be used as an effective method in its quality and production control.
