摘    要:目的]采用反相高效液相色谱法(RP-HPLC)测定文殊兰药材不同部位石蒜碱的含量。方法]采用Agilent Eclipse XDB-C18柱(5μm,250 mm×4.6 mm),甲醇-磷酸盐缓冲液(pH3~4)-三乙胺(8∶92∶0.1)为流动相,流速1.0 ml/min,检测波长为290 nm的色谱条件测定文殊兰不同部位中石蒜碱的含量。结果]石蒜碱进样浓度在0.081 8~0.818 0μg/ml范围内与峰面积呈良好线性关系(r=0.999 9);石蒜碱平均回收率为98.5%,RSD为1.4%。结论]文殊兰鳞茎中的石蒜碱含量最高,所建立的RP-HPLC简便、准确、重复性好,可作为文殊兰药材的质量控制方法。

Abstract:Objective] The aim was to develop an RP-HPLC method for the determination of lycorine in different medicinal portions of Crinum asiaticum.Method] Agilent Eclipse XDB-C18 column(5 μm,250 mm×4.6 mm)was used with methanol-phosphate buffer(pH3~4)-TEA(8∶92∶0.1) as the mobile phase at a flow rate of 1.0 ml/min,the detective wavelength was set at 290 nm.Result] The linear range of lycorine was 0.081 8-0.818 0 μg/ml(r=0.999 9).The average recoveries of lycorine were 98.5% with corresponding RSD 1.4%.Conclusion] Content of lycorine in bulb of Crinum asiaticum is highest.The method is simple and accurate with good repeatability,which can be used for the quality control of Crinum asiaticum.
