摘    要:目的]考察GA对元胡块茎萌发的影响,以及在不定根发生过程中NO的作用。方法]以元胡块茎为材料,在不同温度下用不同浓度的GA处理元胡,考察其对元胡萌发的影响;利用NADPH-黄递酶作为NOS的标志酶,研究NO在不定根发生中的作用。结果]室外低温(4℃)和室内温度(16℃)下,不同浓度的GA对元胡的萌发均有一定的促进作用,但恒温(22℃)不适宜元胡萌发。各温度下,0.6 mg/LGA处理下元胡萌发率较高。元胡不定根发生区域存在有类似动物NOS的酶,且阳性反应集中在根的分生组织中,尤其是在根原基先端的分生组织,遍及整个胞质。结论]GA对元胡萌发具有促进作用,NO在元胡不定根的发生过程具有重要作用。

Abstract:Objective] To investigate effect of GA on germination of Corydalis yanhusuo W.T.Tang,and to discuss influence of NO on adventitious root generation.Method] Using tuber of Corydalis yanhusuo W.T.Tang as material,germination test of Corydalis yanhusuo W.T.Tang treated with GA of different concentration was carried out at different temperature to investigate effect of GA.With NADPH-diaphorase as a marker for NOS,effect of NO on generation of adventitious root was discussed.Result] At 4 and 16 ℃,Corydalis yanhusuo W.T.Tang treated with GA of different concentration was better in germination,while it could not germinate at 22 ℃ due to decayed tuber.First of all,Corydalis yanhusuo W.T.Tang treated with 0.6 mg/L GA at different temperature was highest in germination rate.The NOS-like enzyme mainly existed in the tissue of adventitious roots and the positive reactions mainly centralized in the root meristem,especially in the tip of root anlage.The positive reactions were full of the whole cytoplast.Conclusion] GA played a positive role in the germination of Corydalis yanhusuo W.T.Tang,and NO appeared to playa key role in the generation and development of adventitious root.
