尿激酶原基因cDNA在BHK21细胞中的表达

摘    要:将人尿激酶原(Pro-UK)基因cDNA分别插入到真核表达载体pcDNA3的CMV启动子和pSV2-neo的SV40启动子下游,构建了重组表达质粒pcDNA3-UK和pSV2-UK,分别将这两种表达质粒以脂质体载体法转染BHK21细胞。ELISA检测结果表明,pcDNA3-UK和pSV2-UK在哺乳动物细胞中能够表达,72h表达量分别为28.7ng/ml和13.5ng/ml。

Abstract:Two expression plasmids, pcDNA 3 UK and pSV2 UK, containing the Pro urokinase (Pro UK) cDNA were constructed by inserting the gene into the downstream of the CMV Promoter and the SV40 Promoter, respectively. These two expression plasmids were introduced into Baby Hamster Kidney 21(BHK21) cells with lipofection method. Supernatants collected 72 hours after transfection were assayed for Pro UK by ELISA. It was demonstracted that Pro UK gene expression can be detected and the expressing level of Pro UK were 28.7 ng/ml and 13.5 ng/ml at 72 hours for plasmid pcDNA 3 UK and pSV2 UK, respectively.
