| 摘 要： | 将人尿激酶原（Ｐｒｏ－ＵＫ）基因ｃＤＮＡ分别插入到真核表达载体ｐｃＤＮＡ３的ＣＭＶ启动子和ｐＳＶ２－ｎｅｏ的ＳＶ４０启动子下游，构建了重组表达质粒ｐｃＤＮＡ３－ＵＫ和ｐＳＶ２－ＵＫ，分别将这两种表达质粒以脂质体载体法转染ＢＨＫ２１细胞。ＥＬＩＳＡ检测结果表明，ｐｃＤＮＡ３－ＵＫ和ｐＳＶ２－ＵＫ在哺乳动物细胞中能够表达，７２ｈ表达量分别为２８．７ｎｇ／ｍｌ和１３．５ｎｇ／ｍｌ。

| Abstract: | Two expression plasmids, pcDNA 3 UK and pSV2 UK, containing the Pro urokinase (Pro UK) cDNA were constructed by inserting the gene into the downstream of the CMV Promoter and the SV40 Promoter, respectively. These two expression plasmids were introduced into Baby Hamster Kidney 21(BHK21) cells with lipofection method. Supernatants collected 72 hours after transfection were assayed for Pro UK by ELISA. It was demonstracted that Pro UK gene expression can be detected and the expressing level of Pro UK were 28.7 ng/ml and 13.5 ng/ml at 72 hours for plasmid pcDNA 3 UK and pSV2 UK, respectively. |
