摘    要:为筛选草莓种质常温下离体保存的适宜培养基,本试验以宝交早生(F.ananassaDuch.)和UC5(F.vescaL.)为试材,通过改变培养基中的无机盐浓度、蔗糖浓度、琼脂浓度,设计了9种培养基,将试管苗进行了常温保存,保存期间调查了不同培养基草莓试管苗丛生芽增殖数,根、愈伤、丛芽的生长量,试管苗的长势,叶绿素的含量及存活率等。结果表明:保存过程中试管苗经历了芽增殖、伸长和根生长的过程,生长越快,养分消耗越多,越不利于保存。草莓试管苗保存培养基应选择适宜浓度的营养物质来限制试管苗的生长。本试验中培养基(1/4MS+0.5mg/LBA+15g/L蔗糖+12.5g/L琼脂)是常温下草莓试管苗保存的最适培养基,在此培养基中可保存草莓试管苗11~13个月。

Abstract:The objective of this study was to establish a suitable medium for the conservation of strawberry in vitro at normal temperature. The strawberry plantlets(F. ananassa Duch. and F.vesca L.) were conserved at normal temperature on nine different mediums with different concentrations of the mineral nutrition, agar and sucrose. During the conservation, the number of buds, the growth of plantlet, root and callus, the content of chlorophyll, survival rate and so on were investigated. The results showed that during the conservation, the faster the plantlets grew, the more nutrition was consumed, and the shorter the plantlets were conserved. So the medium for conservation of strawberry in vitro should contain proper concentration of nutrient substance to limit the plantlet growing. In this study, the medium which was1/4 MS, 0.5 mg/L 6-BA, 15 g/L sucrose and 12.5 g/L agar was tested best. The plantlet in vitro could be conserved more than 11 months under this medium at regular temperature.
