酶联免疫测定技术在研究氟虫腈降解中的应用

摘    要:利用单克隆抗体间接竞争ELISA技术检测氟虫腈在青菜和土壤中的降解规律。其在青菜中的降解动态符合一级降解动力学方程C=0.1927e-0.0666t,降解半衰期为10.4d,r=-0.9787;在土壤中的降解符合一级降解动力学方程C=0.0990e-0.0459t,半衰期为15.1d,r=-0.9826。在室内从紫外线、臭氧、pH值、原子辐照4个方面对氟虫腈消解状况进行了分析,发现紫外线照射25h时,氟虫腈降解率最高可达到90%;通入臭氧60min时,氟虫腈降解率最高可达48.5%;溶液pH为12时,氟虫腈在48h时降解率高达88.0%;而辐照处理对氟虫腈的降解效果不明显。

Abstract:Indirect competitive ELISA based on monoclonal antibody was applied to monitor the degradation kinetics of fipronil in vegetable and soil. Degradation kinetics of fipronil in vegetable followed pseudo-first-order kinetics equation C=0.192 7e~ -0.066 6t, and the half-life value was 10.4 d (r=-0.978 7); degradation kinetics of fipronil in soil followed pseudo-first-order kinetics equation C=0.099 0e~ -0.045 9t, and the half-life value was 15.1 d (r=-0.982 6). The immunological assay method was also used to evaluate the effect of UV-light irradiation, ozone supply in aqueous solution, pH value and Co-60 irradiation on the degradation of fipronil in the laboratory. When the sample was irradiated by UV-light for 25 h, supplied with ozone for 60 min, or treated with pH 12 solution for 48 h, the maximum dissipation rate of fipronil was 90%, 48.5% and 88.0% respectively, while Co-60 irradiation had little effect on the degradation of fipronil.
