小麦愈伤组织生长速率与植株再生能力关系的初步研究

摘    要:本文以4个春小麦品种为材料,筛选出适合小麦成熟胚愈伤组织诱导的最佳培养基为MS+2.4-D2mg/L+NAA0.5mg/L+CH200mg/L;用此培养基对10个品种的成熟胚进行离体培养,较系统地比较了愈伤组织生长速率,可再生性愈伤组织诱导率、再生植株频率等指标的基因型间差异。研究结果表明:愈伤组织生长速率与植株再生频率之间存在密切正向相互关系,相关系数为0.78。作者初步认为,前者可作为基因型再生能力大小的一个选择指标。再生能力强的基因型具有较高的愈伤组织生长速率,而且这一生长速率能在继代培养中保持较长时间。

Abstract:Using 4 spring wheat cultivars as experimental materials, it is found that the best medium for wheat mature embryo of 10 spring wheat cultivars is MS + 2.4-D 2mg/L + NAA 0.5mg/L + CH 200mg/L. The mature embryos of 10 spring wheat cultivars are incubated in vitro and systematically compared for several parameters, including callus growth rate (c. g. r. ) , regenerable callus frequency (r. c. f. ) , and plantlet regeneration frequency (p.r.f.) etc. The results show that the differences of the parameters among different cultivars are significant. Besides, thereseems existing a close positive corelation between e.g. r. and p.r. f..So, we presume that the former parameter might act as an indirect index of the latter. The cultivars with higher p.r.f.are also higher in their c.g.r., moreover, the higher c.g.r.could maintain longer time daring subcultures.
