黄珠子草不同部位总黄酮和总酚含量及抑菌活性测定

摘    要:采用紫外分光光度法,NaNO2-Al(NO3)-NaOH和Folin-Ciocalteu比色法测定黄珠子草根、茎、叶、果实4个部位中总黄酮、总酚含量,采用滤纸片法测定了根、茎、叶、果实4个部位乙醇提取物对7种常见病原菌(草莓枯萎病菌、辣椒疫霉病菌、青霉病菌、黄瓜枯萎菌、西瓜枯萎病菌、大肠杆菌、金黄色葡萄球菌)的抑菌活性.叶中总黄酮和总酚含量均最高,分别为31.07 mg·g-1、23.85 mg·g-1,叶中总黄酮含量分别是茎、根及果实的2.86、1.83、1.21倍;叶中总酚分别是茎、根及果实的27.10、22.29、2.52倍.叶中总黄酮、总酚含量均较其他部位高,果实次之,茎最低.在预设质量浓度为40 mg·mL-1时,叶的乙醇提取物对病原菌的抑制效果最好,特别是对黄瓜枯萎病原菌、大肠杆菌和金黄色葡萄球菌,抑制率分别为100%、89.78%、100%,且最小抑菌质量浓度分别为10 mg·mL-1、20 mg·mL-1、10 mg·mL-1.果实部分的抑菌效果稍次于叶部分的,但对黄瓜枯萎病原菌表现出强抑菌性,其抑制率为98.13%.根部分整体抑菌效果较弱,但对黄瓜枯萎病原菌、大肠杆菌有较强的作用,抑制率分别为65.28%,90.04%.而茎基本上没有什么活性.叶和果实的活性成分含量均较高,抑菌活性也较强,药用价值很高,是黄珠子草的主要药用部位.

Abstract:The objective of this research was to clarify the medicinal value and its bioactitves in root, stem, leaf, fruit of Phyllanthus simplex Retz. Ultraviolet-visible spectrophotometric method was used to determine the content of total flavonoids and total phenolics in root, stem, leaf and fruit of Phyllanthus simplex Retz. Filter paper method was used to determine the antibacterial activity of ethanol extracts of different parts from Phyllanthus simplex Retz. on seven pathogens. The content of total flavonoids and total phenolics were the highest in the leaf, and that were 31. 07 mg · g~(-1) and 23. 85 mg · g~(-1), respectively. Total flavonoids in the leaf were stem, root and fruit of times 2. 86, 1. 83, 1. 21. Total phenolics in leaf were the stem, root and fruit 27. 10, 22. 29, 2. 52 times. The content of total flavonoids in fruit was lower than that in leaf, and was lowest in stem. In the set concentration of 40 mg · mL~(-1) , the antibacterial activity of ethanol extract of leaf was the best. Especially for Fu-sarium oxysporum f. sp. Cucumerinum. H, Escherichia coli, Staphylococcus aureas. And the inhibitory rates were 100%, 89. 78%, 100%, respectively. Its MIC was respectively 10 mg · mL~(-1), 2 mg · mL~(-1), 10 mg · mL~(-1). The antibacterial activity of fruit part was not better than that of leaf part, but the antibacterial activity to Fusarium oxysporum f. sp. Cucumerinum. H was high, and the in-hibitory rate was 98. 13%. The antibacterial activity of root was lower , but the antibacterial activity to Fu-sarium oxysporum f. sp. Cucumerinum. H, Escherichia coli were strong. And the inhibitory rates were 65. 28%, 90. 04%, respectively. The content of bioactives in leaf and fruit were high, and the antibacterial activity were strong. The leaf and fruit were the main medicinal part of Phyllanthus simplex Retz.
