怀地黄脱毒试管苗培养条件的优化

摘    要:研究了怀地黄脱毒试管苗在不同pH值、不同光照强度和不同温度下形态指标的差异,以寻求怀地黄脱毒试管苗的最适培养条件。结果表明:适合培养怀地黄脱毒试管苗的pH值在5.8~6.2,该pH值范围内,试管苗生长较健壮,株高和繁殖系数最大,分别为6.66 cm和4.8。最适合培养怀地黄脱毒试管苗的光照强度为2 500 lx,此光照强度下,试管苗生长较健壮,叶色浓绿,株高和繁殖系数分别为5.2 cm和4.6。最适合培养怀地黄脱毒试管苗的环境温度值为30℃,该温度下试管苗生长较健壮,株高和繁殖系数最高,分别为5.8 cm和4.8。因此,适合怀地黄脱毒试管苗生长的pH值为5.8~6.2、光照强度为2 500 lx、温度为30℃。

Abstract:In this paper,the morphological parameters of virus-free plantlets of Rehmannia glutinosa f.Hueichingensis(Chanet Sehih) Hsiao.were studied under different growing environment(pH value,different illumination intensity and different temperature) to find the optimum culture condition.The results were as follows:The optimum pH value was between 5.8 and 6.2,and the plantlets were healthy under pH5.8-6.2,the highest height and propagation coefficient were 6.66 cm and 4.8,respectively.The optimum illumination intensity was about 2 500 lx and plantlets were healthy under this illumination condition,the colour of leaves was deep green,plant height and propagation coefficient was 5.2 cm and 4.6,respectively.The best proper temperature was 30℃,and the plant height and propagation coefficient were 5.8 cm and 4.8,respectively.In a conclusion,the optimum condition for growth of virus-free plantlets of Rehmannia glutinosa f.Hueichingensis(Chanet Sehih) Hsiao.was:pH5.8-6.2,illumination intensity2 500 lx,and the proper temperature 30℃.
