毛叶枣组培快繁技术研究

摘    要:利用组织培养技术对毛叶枣的组培快繁技术进行了研究 .证明 3、4月是 1年内采集外植体的最佳时期 ,在此阶段对外植体进行培养时 ,有效萌芽率最高 .以MS为基本培养基筛选出适合毛叶枣组培各阶段的适宜激素浓度和配比 ,分别为 :启动培养基MS +6 BA 0 8mg L +IBA 0 4mg L ;增殖培养基MS +6 BA 1 2mg L +IBA 0 5mg L ;生根培养基 1 2MS +IBA 3 0mg L .并阐明了继代次数与增殖系数和生根率的关系 ,提出毛叶枣继代次数以 8代左右为宜 ,8代之后应进行生根培养 .

Abstract:The micropropagation of ber ( Ziziphus mauritiana Lam.) was studied through tissue culture.The results show that it is a perfect time for cutting explants from healthy growing plants in March and April. Because in this period it can improve the rate of germination for stem explants culture. The optimum media were screened out through studies on the condition of initiation, proliferation and rooting culture. Main achievements were shown as follows: MS medium supplemented with BA 0 8 mg/L and IBA 0 4 mg/L was suitable for ber initation. MS medium supplemented with BA 1 2 mg/L and IBA 0 5 mg/L was the best for shoots muliplication. But the medium favourable for rooting was 1/2MS basal medium supplemented with IBA 3 0 mg/L. Furthermore, the paper illustrates the relation between generation and ratio of proliferation or rate of rooting. The results show that eight generations are suitable for shoot multiplication. After eight generations rooting culture should be carried out.
