皖北黄牛随机扩增多态DNA研究

摘    要:选取10条随机引物对皖北黄牛进行RAFD分析,从中筛选出3条可得到稳定扩增产物的多态性引物。根据RAPD结果分别计算了两两个体间的带纹相似率、平均带纹相似率、带纹的平均等位基因频率和平均带纹杂合度及遗传多样性指数,作亲缘关系的类平均聚类并绘制了相关关系聚类图。结果表明,皖北黄牛的基因杂合度较高,带纹总的平均等位基因频率、平均杂合度分别为0.55和0.57,群体内存在着较高的遗传变异。

Abstract:Ten random primers were selected and used to conduct the polymorphic DNA analysis of yellow cattle in Northern Anhui province. Three primers that can be used to get stable polymorphic DNA fragments were chosen after screening these primers. From the RAPD analysis data, the band similarity rates and average band similarity rates between two individuals in the sampling population, average band allele frequencies, average band heterozygosis degree, and the Shannon genetic diversity index were calculated to perform the cluster analysis of heterozygosis arithmetic averages and construct the phylogenetic tree. The results showed that the total average band allele frequencies and average band heterozygosis degree were 0. 57 and 0.55, respectively, which indicated the high genetic variance in the herd.
