Abstract:Mice were intraperitoneally injected with cyclophosphamide solution(80 mg·kg-1) to establish immunocompromised mouse model, and then administered Sprassis latifolia polysaccharides(SCPs) by gavage at low dose(LD, 100 mg·kg-1), medium dose(MD, 200 mg·kg-1), and high dose(HD, 400 mg·kg-1), respectively. For each group, hematoxylin-eosin(HE) staining were used to observe morphological changes in mouse cerebral cortex;malondialdehyde(MDA) content was measured, and activities of superoxide dismutase(SOD), catalase(CAT) and glutathione peroxidase(GSH-Px) were determined. ELISA was used to measure 5-hydroxytryptamine(5-HT), tryptophan hydroxylase-2(TPH2), tumor necrosis factor-α(TNF-α), interleukin-1β(IL-1β), interleukin-6(IL-6) and interleukin-10(IL-10) in mouse cerebral cortex;real-time fluorescent quantitative PCR was used to determine mRNA levels of TNF-α, IL-1β, IL-6, IL-10, 5-hydroxytryptamine 2 C receptor gene(5-HT2CR), calmodulin gene(CaM) and brain-derived neurotrophic factor gene(BDNF);Western blot and immunohistochemistry(IHC) were used to determine expression of cyclic AMP responsive element-binding protein(CREB), p-CREB, calmodulin-dependent kinase Ⅱ(CaMK Ⅱ) and p-CaMK Ⅱ protein. The results showed that compared with the model group, the number of cerebral cortical neurons of mice in all three SCPs groups was increased, the intercellular space in all three SCPs groups was reduced, and the cell structure was more complete;both SOD and CAT actitivies were significantly increased in all three SCPs groups, and GSH-Px activity was significantly increased in both MD and HD;5-HT and TPH2 were significantly increased in HD;TNF-α and IL-10 were significantly decreased in both MD and HD, and IL-1β was significantly decreased in all three SCPs groups;mRNA expression of TNF-α, IL-1β, and IL-10 were significantly decreased in all three SCPs groups, and mRNA expression of IL-6 was significantly decreased in HD;mRNA expression of 5-HT2CR and BDNF were significantly increased in both MD and HD, and mRNA expression of CaM was significantly increased in HD;expression of p-CREB was significantly increased in HD, and expression of CaMKⅡ and p-CaMKⅡ were significantly increased in both MD and HD. The above results suggested that SCPs ameliorated cerebral cortex damage of immunocompromised mice through regulating oxidative stress state, inflammatory response, 5-HT secretion level and its down stream signaling pathways.
