64份小麦种质的赤霉素响应评价及矮秆基因分子鉴定

摘    要:为深入了解小麦矮秆种质资源的性状表现和遗传组成,研究了64份小麦品种(系)的株高和节间长度对外施赤霉素的响应,并利用分子标记对矮秆基因组成等进行了鉴定。结果表明,不同材料的株高、节间数量和节间长度对赤霉素响应存在差异。节间长对赤霉素的响应集中在倒二节、倒三节和倒四节。供试材料可以通过增加穗长和穗颈长弥补节间数量减少对株高的影响。赤霉素浓度对供试材料株高构成指数影响较小。通过对不同材料株高构成指数分析发现,SH-04和川麦86两份材料具有良好的株高构成比例。分子标记鉴定表明,供试材料中Rht-B1bRht-D1bRht8的分布频率为31.25%、10.94%和65.63%。矮秆基因的降秆效应表现为Rht-B1b>Rht8Rht9Rht13。本研究得出Rht9+Rht13的株高构成指数为0.613,最接近于0.618,含有该基因组合的材料可以在株型育种中加以利用。

Abstract:In order to further understand the trait performance and genetic composition of wheat dwarf germplasm resources and provide guidance for breeding and utilization, this study evaluated the response of plant height and internode length of 64 wheat varieties(lines) to gibberellin, and identified the dwarf gene composition by molecular markers. The results showed that different materials’ plant height, internode number and internode length had different responses to gibberellin. The response of internode length to gibberellin was concentrated in the second, third and fourth internodes under the spike. The test materials could compensate for the effect of reduced internode number on plant height by increasing the length of spike and peduncle. Different concentrations of gibberellin treatment had little effect on the plant height composition index of the test materials. Analysis of the plant height composition index of different materials found that SH-04 and Chuanmai 86 had good plant height composition ratio. Molecular marker identification showed that the distribution frequencies of Rht-B1b, Rht-D1b, and Rht8 were 31.25%, 10.94%, and 65.63%. Analysis on the dwarf genes found that the dwarfing effect ranked as Rht-B1b>Rht8, Rht9, and Rht13. In this study, the plant height composition index of Rht9+Rht13 was 0.613, which is the closest to 0.618, and the materials carrying this gene combination can be used in plant type breeding.
