金针菇假定蛋白E3泛素连接酶基因的鉴定与表达分析

摘    要:以金针菇基因组与转录组数据为基础,通过本地BLAST方法鉴定金针菇假定蛋白E3泛素连接酶基因,命名为GENE5116,并对其结构、编码蛋白的基本性质与表达模式进行了分析,以期了解其在金针菇子实体生长发育过程中的作用。结果表明:该基因有9个外显子,8个内含子,基因总长为1998 bp,共编码438个氨基酸残基,相对分子质量为48987.36,等电点8.83;系统进化分析结果表明该基因与真菌E3泛素连接酶具有高度同源性,与陶兰柱担菌(Cylindrobasidium torrendii)亲缘关系最近;实时荧光定量PCR结果表明该基因在菌柄的表达量最高且菌柄上部基因表达量显著高于下部,在菌丝的表达量最低,所以推测该基因可能与子实体菌柄伸长有关。

Abstract:Based on the data of genomic and transcriptome data from Flammulina velutipes,the Flammulina filiformis′s putative protein E3 ubiquitin ligase gene was identified,named GENE5116,and its structure was analyzed,basic properties and expression pattern of the encoded protein,for understanding their role in the growth and development of Flammulina velutipes fruit bodies.The results showed that the gene had 9 exons and 8 introns,with a total length of 1 998 bp,encoding a total of 438 amino acid residues,a relative molecular mass of 48 987.36,and an isoelectric point of 8.83.Phylogenetic analysis showed that the gene had a high degree of homology with the fungal E3 ubiquitin ligase and had a closest relationship with Cylindrobasidium torrendii.The results of real-time fluorescence quantitative PCR showed that the gene was most highly expressed in the stalk and the gene expression in the upper part of the stalk was significantly higher than that in the lower part,the gene had the lowest expression in the mycelium.Therefore,it was speculated that the gene might be related to the elongation of the stipe.
