不同龄期柞蚕的表皮生长因子受体(ApEGFR)基因的表达特性分析

摘    要:根据NCBI公共数据库获取的家蚕表皮生长因子受体基因序列,设计8对针对表皮生长因子受体基因的特异性引物。提取柞蚕中不同龄期和不同组织中总RNA,以Oligo-dT为引物,反转录为第一链cDNA,经PCR扩增,荧光定量PCR检测,结果表明表皮生长因子受体基因在不同发育阶段中的表达有差异。不同发育阶段和整个眠期的柞蚕中,ApEGFR的mRNA均有表达,但在5龄和3眠期的时候的表达量最高。不同组织中,在柞蚕的丝腺中ApEGFR的mRNA表达量最高,表皮最低。

Abstract:According to the sequence of silkworm epidermal growth factor receptor gene obtained from NCBI public database,this experiment designed 8 pairs of specific primers for epidermal growth factor receptor gene.Total RNA was extracted from tussah at different ages and in different tissues.Oligo-dT was used as primer and reverse transcribed into first strand cDNA,which was amplified by PCR and detected by fluorescence quantitative PCR.The results showed that the expression of epidermal growth factor receptor gene was different at different developmental stages.ApEGFR mRNA was expressed in tussah at different developmental stages and in the whole molter stage.However,the expression level was highest at the 5 th and 3 rd instars.In different tissues,ApEGFR mRNA expression was highest in tussah silk gland and lowest in epidermis.
