常春藤总DNA提取方法的改进

摘    要:由于常春藤组织内含有较多的酚类和多糖类物质,获得高质量的总DNA有一定难度.现以常春藤叶片为材料,采用不同方法进行常春藤总DNA的提取,并对提取方法进行了改进、优化,有效去除了多糖类杂质的干扰.结果表明:CTAB法、SDS法及改进方法均可进行总DNA的提取,但改进的方法优于常规CTAB法和SDS法,所得的DNA无降解现象,且无杂质污染.

Abstract:Because the phenols and polysaccharides in Ivy are rich,it is difficult to obtain high-quality total DNA from its tissues.In the paper,the total DNA of Ivy was extracted from its leaves,and the different extractingmethods were compared.The extractingmethods of total DNA of ivy were improved and optimized for clearing away polysaccharides effectively.The results showed that the CTABmethod,the SDSmethod,and the improvedmethods can extract total DNA.But the improvedmethods were better than the conventional CTAB and SDSmethod,because they could obtain high-quality total DNA with less elimination and impurity.
