| 摘 要： | 以比活100 IU/mL以上的孕马和孕驴血清为原料,应用HPO2沉淀-超滤浓缩(或用聚 乙二醇浓缩)-乙醇分部沉淀-超滤截留-SE-SephadexC-50柱层析-羟基磷灰石柱层析,进 行PMSG提纯试验,获得了比活9 000 IU/mg左右的高纯度产品,经PAG圆盘电泳只出现一条带.

| Abstract: | A simple and effective procedure was developed for purification of PMSG from pregnant mare or donkey serum whose activity was more than 60IU/mL, The purification procedures included pH fractionation with metaphosphoric acid (HPO3) , concentration with ultrafiltration or poly-ethyleneglycol (PEG), fractionated preciptation with alcohol,ultrafiltration, chromatography on sulfoethyl (SE) -Sephadex G-50 and on hydroxylapa-tite. The specific activities of PMSG in each purification were stepped up to alout 100, 700, 1500, 3000 and 9000 IU/mg and the yield percentages were about 90%, 59%, 97%, 94% and 46%, respectively. The last three purification steps could be able to get rid of miscellaneous proteins note-worthily from crude preparations. The purified preparation showed only one band of PMSG by polyacrylamide gel electrophoresis. |
