甘蓝自交不亲和性的快速测定

摘    要:利用荧光显微镜观察经过处理的甘蓝自花授粉花期柱头和蕾期柱头 ,发现花粉粒在柱头上的萌发及花粉管在柱头表面和花柱中的生长情况有很大差异。自交不亲和系花期柱头有少量的花粉粒萌发 ,并在柱头表面发生严重的胼胝质反应且花粉管的形态不正常 ,要么弯曲、要么背向生长 ,不能穿越乳突细胞壁 ;而自交亲和系花期、蕾期和自交不亲和系蕾期的柱头上 ,花粉粒几乎都能萌发长出花粉管 ,并能穿过花柱到达子房。利用荧光显微镜观察花柱中的花粉管数量可以省时、省工、直观快速鉴定甘蓝自交不亲和性

Abstract:Flower stigmas of different periods after pollination were collected and stained with aniline blue.The degree of pollen tubes in stigma surface and style were observed under fluorescence microscope.The result showed that there were obvious differences in germination of pollen grains and growth of pollen tubes between flower stage and bud stage.There were less attached pollen grains and pollen tube growth in flower stage than in bud stage.The main barrier of self incompatibility in flower stage was that pollen tubes failed to penetrate stigma whose surface produced serious callose reaction,giving rise to the distorting of pollen tubes,swelling of their tops and failing to growth ,but more than fifteen pollen tubes in bud stage were succeed to penetrating stigma whose surface produced little callose reaction,finally getting to ovary along cell space near vascular stand.We can screening fast and truely self incompable and self compatible lines through the method.$$$$
