沙漠肉苁蓉基因组DNA的提取方法比较

摘    要:目的]探索适合肉苁蓉基因组总DNA的提取方法。方法]分别采用改良十六烷基溴化胺(CTAB)法、快速一步(ROSE)法和改良十二烷基苯磺酸钠(SDS)法提取肉苁蓉肉质茎中的基因组DNA,然后通过紫外分光光度计测定不同方法所提取DNA的纯度和浓度,并通过琼脂糖凝胶电泳检测,比较几种提取方法的效果。结果]3种方法所提取的DNA纯度和浓度有差别,从综合的结果来看,以改良的SDS方法为好,可以比较彻底地除去酚类和糖类,可直接用于下一步的研究。结论]改良SDS法为较适合沙漠肉苁蓉基因组DNA提取的方法。

Abstract:Objective] This paper aims to explore genomic DNA of Herba cistanche.Method] Three extraction methods are used including modified CTAB,ROSE and modified SDS.Then,purity and concentration of the genomic DNA are detected by UV spectrophotometer,electrophoresis and restriction enzyme digestion.Several kinds of extraction methods are compared.Result] The purity and concentration of DNA extracted by three methods are different.According to the comprehensive results,the method of modified SDS is best,which can relatively eliminate thorough the phenol and carbohydrate.Conclusion] The modified SDS method fits the desert Cistanche genomic DNA extraction method.
