摘    要:采用平板透明圈法,经初筛和复筛从高黎贡山土样中分离出1株产蛋白酶活性较高的菌株CM8,并对其形态和生理生化特征进行研究。结果表明,CM8菌株的菌落呈不规则形,乳白色,表面湿润,边缘整齐;在光学显微镜下,菌株呈杆状,具运动性,革兰氏染色阳性。经测定,其最适生长温度为32℃,最适生长pH值为7.5,菌株可在酪素平板上产生清晰可见的透明圈,透明圈直径和菌落直径的比值为3.71。菌株产高温蛋白酶的酶活力可达34.24 U/m l。

Study on the Isolation and Physiological Characters of a Producing Strain of Protease

Abstract:Using plate-transparent circle method,a producing strain of high-activity protease,CM8,was isolated from soil sample of Gaoligong Mountain through preliminary and secondary screenings and its shape and physiological and biochemical property were studied.The results indicated that the colony shape of CM8 was irregular and its colony was ivory white and had moist surface and trim edge;under light microscope,the strain was bacillar,mobile and positive in gram staining.The determination results showed that the optimum temperature and pH value for its growth were 32 ℃ and 7.5 resp.The strain could produce clearly visible transparent circles on casein plates,the diameter ratio of transparent circle to colony was 3.71 and the activity of high temperature protease produced by this strain could reach 34.24 U/ml.
