摘    要:[目的]建立多花兰的组培快繁技术体系。[方法]以多花兰种子为试验材料,研究基本培养基和添加物对其无菌萌发、原球茎增殖与分化、试管苗壮苗培育的影响。[结果]在MS、VW、KC、HP[Hyponex(花宝1号)3 g/L+Peptone(蛋白胨)4 g/L]4种培养基中,以VW中的萌发率最高,达到75%;添加AgNO31 mg/L+La(NO3)2.6H2O1 mg/L对原球茎的增殖与分化起促进作用;添加0.3%的活性炭可克服培养过程中的褐化现象;HP培养基添加10%香蕉可以获得壮苗。[结论]在不同培养阶段,通过基本培养基与添加物的优化组合可以建立高效的多花兰无菌播种试管成苗技术体系。

Research on the In vitro Seed Germination of Cymbidium floribundum L and Its Plantlet Formation

Abstract:[Objective] The purpose of this study was to establish a reliable protocol for the propagation of Cymbidium floribundum in vitro.[Method] The effect of the basic medium and other compounds on the in vitro seed germination,protocorm differentiation and plantlet growth of Cymbidium floribundum was studied respectively.[Result] The results indicated that the optimal medium for seed germination was VW,which germination rate was as high as 75%,compared with other three ones(MS,KC and HP).The proliferation and differentiation of protocorm were promoted after 1mg/L of AgNO3+1 mg/L of La(NO3)2·6H2O were added.The protocorm-browning could be alleviated with 0.3%/L of activated charcoal.The strong plantlet could be formed based on the 10%/L of banana juice added in HP.[Conclusion] The high-efficiency system of the Cymbidium floribundum plantlet formation could be established through the optimal combination of basic medium and additives in different culture stages.
