灰葡萄孢菌激活蛋白对番茄灰霉病的诱导抗性及防御相关酶活性的影响

摘    要:为探索激活蛋白对植物诱导抗病性机理,分析诱导过程中植物的应激反应,采用室内盆栽的方法,研究了来源于灰葡萄孢菌(Botrytis cinerea)的14 kD激活蛋白PEBC2诱导番茄对灰霉病的抗性,结果表明,经1.182μg/mL激活蛋白诱导处理后番茄对灰霉病的抗性有显著提高,番茄接种灰霉菌17 d后,对灰霉病的诱抗效果达到67.03%。测定了番茄体内与抗病代谢有关的苯丙氨酸解氨酶(PAL)、过氧化物酶(POD)和多酚氧化酶(PPO)活性的动态变化,经激活蛋白处理后,番茄幼苗叶片的苯丙氨酸解氨酶(PAL)、过氧化物酶(POD)和多酚氧化酶(PPO)活性均有不同程度提高,PAL活性在诱导48 h后达到最高,比对照提高53.82%;POD活性在诱导168 h后达到最高,是对照的2.63倍;PPO活性在24 h和120 h出现2个峰值,分别比对照增加77.57%和87.21%。说明防御相关酶活性的提高,是激活蛋白诱导番茄植株抗灰霉病的主要生理机制之一。

Abstract:A 14 kD protein elicitor from Botrytis cinerea,which can induce tomato to gain resistance of gray mould disease,was researched by the method of pot culture in the green house.The induc- ing efficiency reached 67.03 % at 17th day post inoculation of pathogen when seeds were dipped in 1.182μg/ml activator protein.To reveal the mechanism of resistance induction,the dynamic varia- tion of PAL,POD,PPO which relate with resistance metabolism were measured.The activity of Phenylalanine ammonia lyase(PAL)Peroxides(POD)and polypheol oxidase(PPO)were in- creased effectively after inducement The activity of PAL increased by 53.82 % at 48h.The maxi- mum activity of POD was 2.63 times the control group and occurred at 168 h.The activity of PPO increased by 77.57 % and 87.21% at 24 h and 120 h,respectively.The enhancement of activity of the enzyme correlated with defense is one of the main physiological mechanisms of protein activator inducing disease resistance.
