摘    要:将沙丁胺醇(SAL)经过琥珀酸单酯化后,采用混合酸酐法与载体蛋白偶联,用紫外扫描(UV)和变性凝胶电泳(SDS-PAGE)方法检验偶联效果,推算分子结合比;制备全抗原后免疫BALB/c小鼠,间接ELISA测定多抗血清效价,阻断ELISA鉴定其敏感性。UV和SDS-PAGE分析结果表明,成功制备了BSA-HS-SAL,SAL-HS与BSA的分子结合比为11∶1;ELISA结果表明,获得了高效价、敏感的SAL多克隆抗体,为SAL残留免疫学检测方法的建立奠定了基础。

Abstract:In this study,the mixed anhydride reaction was employed to conjugate salbutamol sulfate(SAL) with large carrier protein.The monosuccinyl derivative of SAL(SAL-HS) was prepared by mixing the SAL hapten with succinic anhydride in a given condition,linked to bovine serum albumin(BSA) and a BSA-HS-SAL conjugation obtained.The immunogen of BSA-HS-SAL was identified by UV spectrogram and SDS-PAGE.BALB/c mice were immunized with BSA-HS-SAL and the titre of polyclonal antibody(pAb) was determined by indirect ELISA.The sensitivity of pAb was identified by blocking ELISA.The results of UV spectrogram and SDS-PAGE showed that the characteristic peak of BSA-HS-SAL has been changed compared to BSA and SAL.The conjugation ratio of SAL to BSA was about 11∶1.pAbs anti-SAL of high titer and sensitivity have been produced by immunizing BALB/c mice,which made it possible to establish immunoassay for detecting SAL residues in feed and animal food.
