薄层色谱法快速检测大蒜低聚果糖

摘    要:建立了薄层色谱法同时快速检测大蒜中7种低聚果糖的方法。用水提、醇沉、醇溶等方法获得大蒜低聚果糖;通过薄层色谱法分析不同展开条件对各聚合度大蒜低聚果糖的分离效果,并且考察了不同上样量、不同硅胶板长度对大蒜低聚果糖分离效果的影响。结果表明:长10 cm的硅胶板在展开系统V正丁醇.V异丙醇.V水.V乙酸=7∶5∶4∶2中,室温下进行4次展开可快速检测大蒜中聚合度1~7的糖组分,斑点清晰、分离度合适。该方法简单、快速、灵敏、重现性好,可用于快速检测低聚糖的组成和含量。

Abstract:An analytical method of thin layer chromatography(TLC) was developed for simultaneous determination of seven oligosaccharides from garlic.The crude oligosaccharide was isolated from fresh garlic with water,then precipitated with ethanol,and extracted again with ethanol.Different developing agents,content and plate length were investigated by TLC.These experimental results demonstrated that DP1~7 of oligosaccharide from garlic were detected and separated successfully by developed four times in 1-butanol/2-propanol/water/acetic acid(7∶5∶4∶2) at room temperature.This method is suitable for detection oligosaccharide’s integrant and content as it is simple,fast,sensitivity and good repeatability.
