白菜型油菜PABP5基因启动子的克隆及表达特性分析

摘    要:本研究中利用基因组步移法从白菜型油菜中克隆到Poly(A)结合蛋白基因PABP5起始密码子上游长588 bp的一段序列,经软件分析预测表明,该序列具有典型的启动子结构,并且具有多个与花药特异性表达相关的顺式作用元件,如TATA box,CAAT box,AGAAA motif,AGGTCA motif,AAACCCTAA motif,GTGA motif等.为了研究该片段的表达特性,本文将克隆到的序列置换pPBI121中的CaMV35S启动子,驱动其下游的GUS基因,构建了植物表达载体,转化拟南芥,获得了转基因植株.组织化学染色表明,在PABP5启动子Ppabp5的驱动下,报告基因GUS在拟南芥的根尖、第一片真叶叶原基以及花药中特异表达.本研究为PABP5基因的进一步功能研究奠定了基础.

Abstract:5'flanking sequence of Brassica rapa Poly(A)binding protein gene PABP5 was cloned by using PCR-based genomic walking method. Results of sequence analysis showed that the cloned sequence contained several pu- tative cis-elements for anther-specific activity, such as TATA box, CAAT box, AGAAA motif, AGGTCA mo- tif, AAACCCTAA motif, GTGA motif, GCGGGAAA motif and GGGCC motif, as well as several motif for ABA responding.
