花生根瘤菌的深层发酵培养研究

摘    要:选择8种培养基进行振荡培养,从中选出YMA、YMA磷酸氢二铵、土壤浸出液、黄豆饼浸出液4种培养基进行深层发酵培养。在发酵条件控制在温度为28℃,pH值为7.0,溶氧量为90%以上的条件下,共培养5 d,定时取样测定其菌数并绘制生长曲线。结果表明,花生根瘤菌在上述4种培养基中的最大生物量分别为12.31亿,15.80亿,30.20亿,53.20亿个/ml;对数期的代时分别为9.112,9.053,9.086,8.933 h;生物量越大,对数期越长。

Abstract:Eight kinds of culture media were designated to carrying on shaking culture,selecting the YMA,the YMA ammonium phosphate dibasic,the extraction of soil,the defatted soybean leaching solution four kinds of culture medium carried on submerged fermentation culture.The fermentation conditions were controled in 28 ℃,pH 7.0,the oxygen quantity dissolved was above 90 %,and cultured on 5 days,samples were taken,bacteria quantity were counted on time and growth curve were determined.The results indicated that the biggest biomass(hundred million/ml) of peanut Rhizobium in four kinds of culture medium was 12.3,15.8,30.2 and 53.2,respectively;The generation time(hour) of logarithmic phase was 9.112,9.053,9.086 and 8.993 respectively.The longer the logarithmic phase was,the bigger the biomass was.The main fermentation condition influence factor and the control method were discussed in the article.
