摘    要:运用RAPD法检测黑麦JNK居群中含有和不含B染色体的基因组DNA的序列差异,并从100个10碱基随机引物中筛选出5个,采用CRED-RA法,检测含有和不含B染色体基因组DNA的甲基化变异。结果表明,含有和不含B染色体的黑麦基因组中均可以发生CmCGG甲基化修饰,但与不含B染色体的基因组相比,含有B染色体的黑麦基因组中发生了更广泛的CmCGG甲基化变异,这可能是B染色体产生表型抑制或基因沉默的一个重要原因。

Abstract:DNA sequence differences in rye genomes(JNK population) with and without B chromosomes were analyzed by random amplified polymorphic DNA(RAPD).5 out of 100 arbitrary 10-mer primers were screened out,following coupled restriction enzyme digestion and random amplification(CRED-RA) of genomic DNA to detect cytosine methylation changes in genomes with and without B chromosomes.The results showed that DNA methylation(CmCGG) was observed in both genomes with and without B chromosomes.Moreover,genomes with B chromosomes caused extensive DNA methylation(CmCGG) comparing with those without B chromosomes.These epigenetic changes might play a key role in the phenotypic suppression or gene silencing caused by B chromosome.
