青花菜种质资源遗传多样性的ISSR分析

摘    要:利用ISSR分子标记技术,对来自国内外的33个青花菜(Brassica oleracea var. italica)材料进行遗传多样性研究。从100条通用ISSR引物中筛选出15条用于PCR扩增,共得到可重复的211条清晰谱带,其中多态性谱带180条,多态性条带比率(PPB)为8531%,平均Neis基因多样性指数与Shannons信息指数为0332 7和0487 7;材料间遗传距离范围为009~045,平均028;聚类分析表明,33份青花菜种质可分为4组,没有明显的地域分布规律。结果表明,ISSR标记可用于青花菜种质的分子鉴定;青花菜供试材料具有一定的遗传多样性。

Abstract: The genetic diversity of 33 broccoli (Brassica oleracea var. italica) germplasm resources, from home and abroad, was investigated using ISSR molecular marker method. Fifteen primers were selected from 100 universal ones, and they produced 211 distinctive bands, including 180 polymorphic sites with 8531% of polymorphic bands. The average Neis genetic diversity index and Shannons information index were 0332 7 and 0487 7, respectively. The genetic distance ranged from 009 to 045 with an average value of 028. Cluster analysis divided the 33 broccoli materials into 4 groups, and showed no obvious correlation with geographic distribution. The results revealed that the ISSR markers could be used to identify broccoli germplasm resources, and the materials tested showed a certain level of genetic diversity.
