墨西哥菜用仙人掌组织培养初报

摘    要:以墨西哥菜用仙人掌栽培种米邦塔(milpa alta)茎片切块为外植体,研究了茎片年龄、表面消毒方法、植物生长调节剂和活性炭等对其侧芽萌发的影响。结果表明,幼茎片较成熟茎片易于表面消毒,且以70%酒精消毒30s,0.1%升汞杀菌8min效果最佳;与不同质量浓度的6-BA组合,低质量浓度NAA(0.05mg/L)较高质量浓度NAA(0.1和0.5mg/L)促进侧芽提早萌发,培养12d侧芽萌发率最高可达51%(6-BA,1mg/L);0.5%活性炭强烈抑制侧芽的萌发,侧芽萌发率仅1%左右.侧芽在1/2MS NAA0.5mg/L生根培养基上生根,移栽后成活。

Abstract:The effects of method for sterilization,age of stem,plant hormones and active charcoal on bud initiation in Opuntia dinenii cv.milpa alta were studied.It was showed that the young stem was easy to be sterilized,the suitable method for sterilization was 70% ethanol for 30 s,then 0.1% HgCl 2 for 8 min.And in combination with 6 BA,low concentration(0.05 mg/L)of NAA enhanced bud initiation,the highest percentage of bud initiation reached 51% with 6 BA at 1 mg/L.Active charcoal(0.5%)seriously inhibited bud initiation and the percentage reached only 1%.1/2 MS medium containing 0.5 mg/L NAA was optimal for the shoot rooting.
