高粱DNA导入小麦"矮早8"引起后代变异的研究及SSR分析

摘    要:将高梁“抗5”DNA通过花粉管通道导入小麦品种“矮早8”,其后代在株高、蜡质层、芒型、穗型、旗叶、成熟期等几个方面发生大量变异。通过对变异后代系统选择得到多个稳定的变异种质系;通过对“矮早8”、D8—1、D8—4、高粱“抗5”基因组DNA的SSR分子标记检测,引物xgwm6、xgwm148、xgwm295、xgwm328、xgwm257检测出了较丰富的多态性DNA片段.其中xgwm6、xgwm257检出了高粱“抗5”DNA特有而“矮早8”没有的特异带型,进一步说明了D8—1、D8—4是由高粱“抗5”DNA片段嵌入“矮早8”基因组DNA变异而来。

Abstract:There are many descendants had been mutated in plant height.waxiness .awn .Ear type.Flag leave .maturation by importing broomcorn DNA of "kang5" into the wheat "Aizao8" through the Pollen- tube Pathway~ We have own many unaltered germplasm line by Subsequent continuous selection in the variational descendants ; Detect the DNA of" aizao8" D8 - 1 .D8 -4.broomcorn" kang5" Using SSR markers, There are many differential fragments have been found by the Prime xgwm6 ,xgwm148 、 xgwm295, xgwm328, xgwm257, Especially xgwm6, xxgwm257, they have been found the differential fragments in the DNA of D8 - 1 and D8 - 4 that belong to the hroomcorn DNA of "kangS" only, It further attest that D8 - 1 and D8 -4 was come from "AizaoS'" that was inlaid the fragments of broomcorn DNA of "kang5 ".
