Molecular Quantitative Detection for Overwintering of Uromyces striatus in Inner Mongolia
Abstract:Alfalfa is a legume forage and is the most widely grown forage crop in the world,the planting area and yield of commercial alfalfa forage in Inner Mongolia rank the second in China.In recent years, the occurrence of alfalfa rust is becoming more and more serious,the development of alfalfa industry in Inner Mongolia is greatly affected.From 2018 to 2020, the real-time quantitative PCR(qPCR) technology was established based on RNA level to detect the overwintering inoculum quantity of Uromyces striatus in Ulanqab, Hohhot, Baotou, Ordos and Bayannuthe, which are the main alfalfa producing areas in Inner Mongolia.The results showed that the mean molecular disease index(MDI) of overwintering U.striatus was 0.139, 0.284, 0.196, 0.569 and 0.280, respectively.The mean MDI of Ordos city was significantly higher than that of the other four areas (P<0.001).The mean MDI of overwintering U.striatus was 0.43 in 2019, which was significantly higher than that of the other years in 2018(0.24, P<0.05) and in 2020(0.193, P<0.05).U.striatus could overwinter underground rhizomatous tissue of alfalfa.There was a positive correlation between the amounts of overwintering U.striatus and the disease incidence of alfalfa rust after plants regreening (r=0.806, P<0.001).
