二氢杨梅素在小鼠体内的药代动力学研究

摘    要:采用小鼠灌胃和尾静脉注射方式给药后,不同时间点摘眼球取血,HPLC测定小鼠血浆中二氢杨梅素的含量,运用DAS3.2.1软件计算药代动力学参数。建立小鼠血浆中二氢杨梅素含量的HPLC测定方法,并考察其在小鼠体内的动力学过程。所得回归方程相关系数r=0.999 4,最低检测浓度为0.04μg/mL,日内及日间RSD分别为1.9%~4.8%和2.4%~4.4%,准确度RE为-0.8%~7.2%,方法回收率为74.3%~85.9%。二氢杨梅素的血药浓度-时间曲线符合一级吸收一级消除的一房室模型。说明所建立的方法符合生物样品分析指导原则的方法学要求,能够满足二氢杨梅素在小鼠体内的药动学研究要求。

Abstract:To establish a method for the determination of dihydromyricetin in mice serum and investigate the pharmacokinetic characteristics of dihydromyrieetin in mice, the blood samples were collected at different time points from male mice treated with dihydromyricetin, and dihydromyricetin contents were measured by HPLC system. The correlation coefficient of the regression equation was 0. 999 4 and the limit of detection was 0. 04 μg/mL. The relative standard deviation of intra - day and inter - day were 1.9% - 4. 8% and 2.4% 4. 4% , respectively. The relative error of the accuracy was - 0. 8% - 7.2%. The average recoveries of dihydromyricetin were 74.3% - 85.9%. The serum concentration - time curve of dihydromyricetin was better consistent to a one - compartment model with first order absorption and first order elimination, therefore, the established method may satisfy the requirement for DMY pharmacokinetic research.
