千层金离体组织培养与诱导再生研究

摘    要:为了研究千层金快速繁殖技术和品种的基因工程改良,本研究以千层金叶片为外植体,研究了不同激素浓度对千层金叶片再生的影响。结果表明:最佳愈伤组织诱导培养基为MS+TDZ 0.01 mg/L+NAA 0.1 mg/L,诱导率可达76.05%;最佳分化培养基为MS+TDZ 0.02 mg/L+NAA 0.05 mg/L,愈伤分化率可达67.13%;最佳生根培养基为1/2MS+IBA 0.2mg/L,生根率最高为71.27%。

The In Vitro Culture and Plant Regeneration of Melaleuca bracteata via Organogenesis

Abstract:In order to establish the regeneration system for micropropagation and genetic engineering of Melaleuca bracteatais, the effects of hormone concentrations on the regeneration efficiency were investigated using Melaleuca bracteatais leaves as the explants. The results showed that the best medium for callus induction was the MS containing 0.01 mg/L TDZ and 0.1 mg/L NAA, by which the callus induction frequency was as high as 76.05% . The best shoot regeneration rate (67.13%) was obtained from MS medium supplemented with 0.05 mg/L NAA and 0.02 mg/L TDZ. The 1/2MS added with 0.2 mg/L IBA, 3% sucrose and 0.5% agar gave the highest rooting rate (71.27%).
