农杆菌素MI15的分离、纯化及分子量测定

摘    要:放射土壤杆菌(Agrobacterium radiobacter)MI15菌株.属于生物Ⅰ型.在平皿培养时,能够产生农杆菌素MI15,能抑制葡萄根癌土壤杆菌的生长.采用透析和DEAE纤维素柱离子交换层析纯化制备了MI15菌株分泌的农杆菌素MI15.经HPLC分析,此制备物纯度较高,可达到结构分析要求.紫外吸收光谱分析显示,该物质在260nm和280nm处均无洗脱峰,故认为该物质不是核酸、蛋白类似物.经红外吸收和元素分析,初步认定它是一种小分子有机物,质谱测定结果显示,农杆菌素MI15的分子量为368.

Abstract:The MI15 strain of Agrobacterium radiobacter can produce Agrocin MI15 which inhibits the growth of A grobactium tumefacwns,a pathogen causing grape crown gall. Agrocin MI 15 was purified with dialysis followed by DEAE-cellulose column chromatography. HPLC analysis showed that a pure Agrocin MI 15 was obtained. UV spectrophotometric analysis indicated that the Agrocin MI 15 was not nucleic acid or protein-like substance because Agrocin MI 15 had no absorption spectrum at 260nm and 280nm. IR spectroscopy and element analysis proved Agrocin MI 15 to be a small molecular organic sub-strance. MS analysis indicated that the molecular weight of Agrocin MI 15 was 368.
