不同溶剂提取的钩吻粗提物中成分含量的测定

钩吻为马钱科钩吻属常绿藤本植物,具有抗肿瘤、免疫调节、促生长等功效,研究表明其主要活性和毒性成分均为吲哚类生物碱。钩吻的药效剂量与中毒剂量十分接近,且致死量低,为给钩吻广泛的临床应用提供理论依据,分别采用酸性染料比色法、苯酚-硫酸法、考马斯亮蓝法对3种不同溶剂工业化提取的钩吻粗取物中生物碱、多糖、蛋白质进行含量测定。结果显示,钩吻醇提(950 mL/L乙醇)、酸水提(5 mL/L H2SO4)、水提物中生物总碱含量分别为64.4g/L、21.5g/L和5.7g/L,糖含量分别为264.1g/L、89.6g/L和9.3g/L,蛋白含量分别为44.7g/L、12.7g/L和34.6g/L。

Content Determination of Ingredients in Three Different Crude Extracts of Gelsemium elegans

Gelsemium elegans is an evergreen liana of Loganiaceae,which has anti-tumor,immunomodula-tion,promoting growth and other effects.Research showed that the main active and toxic ingredients are indole alkaloids.The efficacy dose and toxic dose of Gelsemium are very close,and lethal dose is low.To provide a theoretical basis for a wide range of clinical applications to Gelsemium,acid dye colorimetry,phe-nol-sulfuric acid method,Coomassie brilliant blue method were respectively used to determine alkaloids, polysaccharides,proteins of three crude extracts from three different extraction solvents in this study.The results showed that total alkaloids contents of ethanol extract (95% ethanol),acid extract (0.5% H 2 SO4 ), water extract of Gelsemium were 6.44%,2.15%,0.57%,sugar contents were 26.41%,8.96%,0.93%, protein contents were 5.91%,1.36%,4.23% ,respectively.
