白藜芦醇对氧化应激仔猪肝脏抗氧化酶、细胞因子mRNA表达及核因子E2相关因子2相关因子信号通路的影响

摘    要:本试验旨在研究白藜芦醇(RES)对敌草快(diquat)诱导的氧化应激仔猪肝脏抗氧化、抗炎性能的影响。选用健康断奶仔猪30头,随机分为5组,对照组(CON组)和diquat组(DIQ组)饲喂基础饲粮,试验组(RES-10组、RES-30组、RES-90组)饲喂在基础饲粮中分别添加10、30、90 mg/kg RES的试验饲粮。于试验第15天清晨给DIQ组、RES-10组、RES-30组、RES-90组仔猪按照10 mg/kg BW剂量腹腔注射diquat建立氧化应激模型,CON组注射相同剂量灭菌生理盐水。试验期21 d。结果表明:与CON组相比,DIQ组肝脏超氧化物歧化酶1(SOD1)、超氧化物歧化酶2(SOD2)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶-1(GPX-1)的mRNA相对表达量均显著降低(P<0.05);肝脏白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)的mRNA相对表达量显著升高(P<0.05),而白细胞介素-10(IL-10)的mRNA相对表达量显著降低(P<0.05);肝脏Kelch样ECH关联蛋白1(Keap1)的mRNA相对表达量显著升高(P<0.05),核因子E2相关因子2(Nrf2)和醌氧化还原酶1(NQO1)的mRNA相对表达量显著降低(P<0.05);肝脏Keap1和Nrf2蛋白表达量分别上升和下降,NQO1的蛋白表达量则呈现显著下降(P<0.05)。与DIQ组相比,RES-10组、RES-30组、RES-90组肝脏SOD1、SOD2、CAT、GPX-1的mRNA相对表达量均显著上调(P <0.05);RES-30组、RES-90组肝脏TNF-α、IL-1β、IL-6的mRNA相对表达量均显著下调(P<0.05),同时肝脏IL-10的mRNA相对表达量显著上调(P<0.05),且RES-90组效果最显著;RES-10组、RES-30组、RES-90组肝脏Keap1的mRNA相对表达量均显著下调(P<0.05),同时肝脏Nrf2、NQO1的mRNA相对表达量显著上调(P<0.05),且RES-90组效果最优;肝脏Keap1蛋白表达量在RES-90组显著下降(P<0.05),Nrf2蛋白表达量在RES-90组有上升的趋势(P=0.057)。综上所述,RES可以提高氧化应激仔猪肝脏抗氧化、抗炎性能,降低肝脏氧化应激程度,其发挥抗氧化机制可能与Nrf2信号通路有关。

Abstract:The purpose of this experiment was to study resveratrol(Res)on the liver anti-oxidative and antiinflammatory capacity in diquat challenged oxidative stress piglets. Thirty weaned piglets with good health were selected and randomly divided into 5 groups,the control group(CON group)and diquat group(DIQ group)was fed a basal diet,RES of 10,30 and 90 mg/kg were added to the basal diet in the experimental groups(RES-10 group,RES-30 group,RES-90 group),respectively. In the early morning of the 15 th day of the experiment,the piglets in the DIQ group,RES-10 group,RES-30 group and RES-90 group were intraperitoneally injected with diquat at a dose of 10 mg/kg BW to establish an oxidative stress model,and the control group injected the same dose of sterilized saline as a reference. The trial period was 21 days. The results showed as follows:compared with the CON group,the liver superoxide dismutase 1(SOD1),superoxide dismutase 2(SOD2),catalase(CAT)and glutathione peroxidase-1(GPX-1)mRNA relative expression levels were all significantly decreased in the DIQ group(P<0.05);the liver interleukin-1β (IL-1β),tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)mRNA relative expression levels were significantly increased(P<0.05),while the liver interleukin-10(IL-10)mRNA relative expression level was significantly reduced(P<0.05);the liver Kelch-like ECH-associated protein 1(Keap1)mRNA relative expression level was significantly increased(P<0.05),while the liver nuclear factor erythroid 2 related factor 2(Nrf2)and NADPH quinone oxidoreductase 1(NQO1)mRNA relative expression levels were significantly reduced(P < 0. 05);the liver Keap1 and Nrf2 protein expression levels were increased and decreased,respectively,and the NQO1 protein expression level was reduced significantly(P<0.05). Compared with the DIQ group,SOD1,SOD2,CAT and GPX-1 mRNA relative expression level was significantly increased in the RES-10 group,RES-30 group and RES-90 group(P<0.05);in the RES-30 group and RES-90 group,liver TNF-α,IL-1β and IL-6 mRNA relative expression levels were significantly reduced(P<0.05),meanwhile,IL-10 mRNA relative expression level was significantly increased(P<0.05),among which the RES-90 group had the most significant effect;liver Keap1 mRNA expression level was significantly dropped in the RES-10 group,RES-30 group and RES-90 group(P<0.05),while the Nrf2 and NQO1 mRNA relative expression levels were significantly increased(P<0.05),among which the RES-90 group had the best effect;the liver Keap1 protein relative expression level was significantly decreased in the RES-90 group(P<0.05),and the Nrf2 protein expression level was tend to increase in RES-90 group(P = 0.057). It is concluded that RES increase the anti-oxidative and anti-inflammatory capacity in the liver of oxidative stress piglets,and reduce the level of liver oxidative stress,its anti-oxidative mechanism may be related to Nrf2 signaling pathway.Chinese Journal of Animal Nutrition,2022,34(2):1230-1239]
