亚洲玉米螟幼虫P450基因片断的克隆与序列分析

摘    要:根据G enB ank中昆虫P 450氨基酸保守序列设计了1对简并引物,以亚洲玉米螟田间自然种群基因组DNA为模板,扩增得到一条长度为603 bp的特异性DNA片段,编码201个氨基酸。将此氨基酸序列与G en-B ank中已知序列采用B last及C lusta lW EB I软件进行同源性分析,结果表明该氨基酸序列与CYP 4家族的同源性较高,达37%~53%,初步推测此序列为CYP 4家族的一员。

Abstract:With genomic DNA of Ostrinia furnacalis field natural population as the template,and a pair of degenerate primers were designed according to the conservative amino acid sequences of insect P450 from GenBank,a specific DNA fragment with 603 bp was obtained,encoding 201 amino acid residues. Homologous analysis was done on the deduced amino acid sequence to GenBank through Blast and ClustalW EBI software,and the results demonstrated that the sequence had as high as 37 %--53% identity to members of CYP4 family,and suggested that the sequence belongs to CYP4 family.
