摘    要: 本文报道利用依主梨树茎尖、侧芽进行离体培养的实验过程.春、秋季取成年依主梨树一年生枝条的顶芽或侧芽作实验材料,其中秋季芽先用赤霉素打破休眠,经剥离、消毒后接种于诱导生长和分化的培养基上,生长约一个月后转入增殖培养基,40天后将增殖后伸长的芽条用于生根,其余小芽则继续增殖;生根的梨苗则进行移栽.通过实验,MS/2(大量元素)+MS(微量元素)+铁盐+有机成分(简称培养基A),附加6-BA 2mg/L、NAA 0.2mg/L、GA3 1~3mg/L适于芽生长和分化;而附加6-BA 1.5mg/L、NAA 0.2mg/L、QA3 2mg/L是优良增殖培养基;附加IBA 10mg/L处理8天的二步生根法较好;生根的试管苗已移栽定植成活.

Abstract:One year's terminalor axillary buds the adult Yizhu pear were used. in spring and autumn. The autumn buds were prebroken dormance by GA3, after stripped and sterilized, the buds were inoculated in the medium of growth and differentiation for one month, then transfered them into medium of multiplication for 40 days, the longer shoots were used for rooting, and then the rooting shoots were planted into soil. By experiment, the medium added with MS/2 (macroelement), MS (microelement), iron salts and organic substance (ie: Medium A) with agar 6.5%, source 3%, 6-BA 2mg/L, NAA 0.2mg/L and GA3 1~3mg/L was suitable for shoots growth and differentiation, Medium A with 6-BA 1.5mg/L, NAA 0.2mg/L and GA3 2mg/L was a good medium for multiplying buds, Medium A with IBA 10mg/L deal with the young plant for 8 days, then transfered them into Medium B [MA/2 (macroelement), MS (microelement), iron salts] without hormone, this two steps rooting method is better than that of one step rooting.
