摘    要:[目的]研究高压静电场对紫花苜蓿组织培养的影响.[方法]利用电场处理紫花苜蓿的子叶和下胚轴,采用正交试验设计,设定3个水平,研究电场对外植体出愈率的影响.[结果]诱导愈伤组织的最佳浓度组合是1.8 mg/L2,4-D +0.3 mg/L 6-BA;下胚轴在培养7d后出愈率最高务件为频率3.85 kHz,场强2 kV/cm,时间20 min;20 d后,由于用20 min处理的下胚轴逐渐凋亡,因此下胚轴出愈率最高的条件为频率3.85 kHz,场强2 kV/cm,时间15 min.试验发现经电场处理后外植体木质化程度较高.[结论]高压静电场对紫花苜蓿组织培养具有一定影响.

The Effect of Electric Field on Alfalfa Tissue Culture

Abstract:[Objective] To study effects of electric field on alfalfa tissue culture.[Method] The cotyledon and hypocotyl of alfalfa is treated with electric field combined with tissue culture technology.We adopted the orthogonal experimental design and set up three levels,in order to study the influence of explant callus induction rate.[Result] The results indicated that the optimal concentration combination of callus induction is 2,4-D:1.8 mg/L,6-BA:0.3 mg/L; the condition of the highest callus induction rate is frequency 3.85 kHz,field strength 2 kV/cm,time 20min after 7 days cultivation of hypocotyl.After 20 days,20min treatment has promoted hypocotyl apoptosis,therefore the condition of the highest hypocotyl callus induction rate is frequency 3.85 kHz,field strength 2 kV/cm,time 15 min.In addition,we found that explant lignification degree is higher after electric field treatment.[Conclusion] Electric field has a certain effect on tissue culture of alfalfa.
