利用近等基因系定位玉米无叶舌基因的研究

摘    要:2008年发现隐性遗传的无叶舌突变株,经过连续自交后育成无叶舌自交系黄早四lg。为了充分挖掘并利用玉米无叶舌优异的基因资源,分别以5个玉米骨干自交系为轮回亲本、黄早四lg为供体亲本进行回交转育,获得相应的无叶舌自交系吉V203lg、K10lg、吉V088lg、吉V152lg和哲461lg,同时获得5对玉米无叶舌性状的近等基因系。以5对近等基因系为定位群体,利用Affymetrix 5H90K基因芯片对5对玉米无叶舌近等基因系进行基因型检测,对玉米无叶舌基因进行定位,在此基础上对候选基因进行初步分析。结果表明,玉米无叶舌基因位于2.01~2.02 bin上约1.12 Mb的物理区间内,在此区间内有64个基因,其中,有26个基因编码功能蛋白,有38个未知功能的蛋白和假定蛋白。

Abstract:A recessive liguleless mutant was found in the self- crossing population of a maize inbred line Huangzao4 in 2008, and then a liguleless inbred line Huangzao4lg was gained through continual self-crossing. In order to explore and make full use of the elite genetic resources of maize liguleless trait, five liguleless maize inbred lines were obtained by back-crossing the inbred lines(JiV203, K10, JiV088, JiV152 and Zhe461) as recurrent parents to Huangzao4lg as donor parent, respectively. That is, 5 near isogenic lines of maize liguleless trait were obtained, too. Five pairs of near-isogenic lines were used as the mapping population, which was genotype by Affymetrix 5H90K SNP chip, and then the gene of the liguleless trait was mapped. In addition, candidate genes were primarily analyzed. The results showed that the maize liguleless gene was in the physical region of about1.12 Mb on the 2.01-2.02 bins, and there are 64 genes totally in this interval, of which 26 genes encoded functional proteins and 38 genes encoded putative proteins and unknown proteins.
