苦瓜总皂甙的提取及含量测定

摘    要:用乙醇浸提热回流法、微孔技术和大孔树脂吸附纯化新工艺提取苦瓜总皂甙。以β-谷甾醇作为标准样,在450~700 nm波长下扫描,吸收光谱表明在545 nm处有最大吸收峰;采用分光光度法对所提取的苦瓜总皂甙进行定量分析。结果表明:苦瓜干粉中总皂甙提取率为0.047 4%,纯度为81.71%;回归方程A=0.002 4X+0.014 8(R2=0.998 3),加样平均回收率为100.90%,RSD为1.32%(n=5)。该研究提取工艺简单、无污染、产品纯度高、可操作性强。分光光度法测定苦瓜总皂甙含量简便、快捷、准确,可作为苦瓜原料及其制品质量评价的方法。

Abstract:The total saponin of balsam pear(Momordica charantia L.) was extracted by the method of alcohol-hot-extraction-reflux,microaperture technique and macroaperture-resin-absorption-newly-purified-technique in this study.Total saponin was scanned(450~700 nm wavelength) and β-sitosterol be used as standard sample and absorption spectrum showed that the peak absorption was located in 545 nm wavelength.Total saponin extracted was quantitatively analysed by spectrophotometry.The experimental results demonstrated that the extraction rate of total saponin of dry powder of balsam pear was 0.047 4%,purity 81.71%,regression equation presented as A=0.002 4X+0.014 8(R2=0.998 3),average recovery rate was 100.90% and RSD(relative standard deviation)1.32%(n=5).The extraction technology was simple,non-pollution,high purity of product and high operating.The determination of content of total saponin of balsam pear by spectrophotometry was simple and convenient,quick and accuracy and it was acted as a method of quality assessment of raw material of balsam pear and its manufactures
