桑叶中乙醇酸氧化酶活性的研究

摘    要:桑叶中乙醇酸氧化酶活性是筛选低光呼吸桑品种的重要指标。采用测定乙醛酸苯腙(GPH)变化量的方法,对不同品种桑叶及用Na2S和草酸处理后桑叶中的乙醇酸氧化酶(GO)活性进行测定的结果表明:不同品种桑叶的GO活性存在显著差异,最大差异近3倍;用Na2S处理桑叶可以提高乙醇酸氧化酶活性,0.5~2.0 mmol/LNa2S处理可使GO活性比对照升高100%~130%;用草酸处理桑叶可以降低乙醇酸氧化酶活性,50~200 mmol/L草酸处理可使GO活性比对照下降49.6%~98.9%。

Abstract:The activity of glycolate oxidase in mulberry leaves is an important index for selection of varieties with low photorespiration.Glycolate oxidase was purified from leaves of different mulberry varieties,and from mulberry leaves treated with sodium sulfide(Na2S) and oxalic acid(C2H2O4),and the activities of glycolate oxidase were determined by the method for testing of glyoxylate phenylhydrazone The results showed that the activities of glycolate oxidase in different mulberry varieties were markedly different;glycolate oxidase activity increased 100%~130% in the mulberry leaves treated with 0.5~2.0 mmol/L Na2S and decreased 49.6%~98.9% in the mulberry leaves treated with 50~200 mmol/L C2H2O4 than the control,respectively.
