小麦超氧物歧化酶的类型及分布

摘    要:用不连续聚丙烯酰胺凝胶电泳法(PAGE)研究了5个小麦品种(系)叶、根,线粒体中超氧物歧化酶(SOD)的类型。不同品种黄化苗的叶和根中都可检测出3条酶活带,其中迁移率最低的是锰SOD,活性占总活性的2.20%~7.20%,铜锌SOD有2条带,居中的带活性占71.1%~81.2%,迁移得最快的带占12.5%~23.7%;旗叶中都可检测出5条带,即在相对于黄化苗SOD电泳图谱上居中的酶活区可分辨3条带;线粒体SOD以锰SOD为主,胞浆中能检测出铜锌SOD。c带相对活性与叶绿体含量有关。将旗叶SOD的5条酶带分别指定为a。b_l,b_2,b_3和c带。

Abstract:SOD types in leaf, root and mitochodrium of 5 wheat cultivars(lines) were studied with discontinuous PAGE. Three SOD activity bands weredetected in shoot and root of etiolated seedlings of all the 5 CVS., of which theslowest migrating band was found to be mangano-SOD constituting 2.20%~7.20% of the total soluble SOD; the fastest migrating band was cupro-zinc SODresponsible for 12.5%~23.7%:the other one was also cupro-zinc SOD making71.1%~81.2%.However, 3 bands were resolved in the intermediate activity zoneof the SOD profile in the flag leaves. Mangano-SOD was predominent in themitochondrium from two cultivars.Only cupro-zinc SOD bands were detectedin the cytosol. The 5 SOD bands are designated as a, b_1, b_2, b_3, and C respce-tively.
